A very simple, re-executable neuroimaging publication.

Reproducible research is a key element of the scientific process. Re-executability of neuroimaging workflows that lead to the conclusions arrived at in the literature has not yet been sufficiently addressed and adopted by the neuroimaging community. In this paper, we document a set of procedures, which include supplemental additions to a manuscript, that unambiguously define the data, workflow, execution environment and results of a neuroimaging analysis, in order to generate a verifiable re-executable publication. Re-executability provides a starting point for examination of the generalizability and reproducibility of a given finding.

Search for dark matter at [Formula: see text] in final states containing an energetic photon and large missing transverse momentum with the ATLAS detector.

Results of a search for physics beyond the Standard Model in events containing an energetic photon and large missing transverse momentum with the ATLAS detector at the Large Hadron Collider are reported. As the number of events observed in data, corresponding to an integrated luminosity of 36.1 fb[Formula: see text]  of proton-proton collisions at a centre-of-mass energy of [Formula: see text], is in agreement with the Standard Model expectations, model-independent limits are set on the fiducial cross section for the production of events in this final state. Exclusion limits are also placed in models where dark-matter candidates are pair-produced. For dark-matter production via an axial-vector or a vector mediator in the s-channel, this search excludes mediator masses below 750-[Formula: see text] for dark-matter candidate masses below 230-[Formula: see text] at 95% confidence level, depending on the couplings. In an effective theory of dark-matter production, the limits restrict the value of the suppression scale [Formula: see text] to be above [Formula: see text] at 95% confidence level. A limit is also reported on the production of a high-mass scalar resonance by processes beyond the Standard Model, in which the resonance decays to [Formula: see text] and the Z boson subsequently decays into neutrinos.

Elastic 'tethers' connect separating anaphase chromosomes in a broad range of animal cells.

We describe the general occurrence in animal cells of elastic components ("tethers") that connect individual chromosomes moving to opposite poles during anaphase. Tethers, originally described in crane-fly spermatocytes, exert force on chromosome arms opposite to the direction the anaphase chromosomes move. We show that they exist in a broad range of animal cells. Thus tethers are previously unrecognised components of general mitotic mechanisms that exert force on chromosomes and they need to be accounted for in general models of mitosis in terms of forces on chromosomes and in terms of what their roles might be.

Autoimmune Subepidermal Bullous Diseases of the Skin and Mucosae: Clinical Features, Diagnosis, and Management.

Autoimmune subepidermal blistering diseases of the skin and mucosae constitute a large group of sometimes devastating diseases, encompassing bullous pemphigoid, gestational pemphigoid, mucous membrane pemphigoid, epidermolysis bullosa acquisita, and anti-p200 pemphigoid. Their clinical presentation is polymorphic. These autoimmune blistering diseases are associated with autoantibodies that target distinct components of the basement membrane zone of stratified epithelia. These autoantigens represent structural proteins important for maintenance of dermo-epidermal integrity. Bullous pemphigoid (BP) is the most common subepidermal autoimmune blistering disease of the skin and mucosae. Although the disease typically presents with a generalized blistering eruption associated with itch, atypical variants with either localized bullous lesions or "non-bullous" presentations are observed in approximately 20% of patients. A peculiar form of BP typically associated with pregnancy is pemphigoid gestationis. In anti-p200 pemphigoid, patients present with tense blisters on erythematosus or normal skin resembling BP, with a predilection for acral surfaces. These patients have antibodies targeting the 200-kDa basement membrane protein. Epidermolysis bullosa is a rare autoimmune blistering disease associated with autoantibodies against type VII collagen that can have several phenotypes including a classical form mimicking dystrophic epidermolysis bullosa, an inflammatory presentation mimicking BP, or mucous membrane pemphigoid-like lesions. Mucous membrane pemphigoid (MMP) is the term agreed upon by international consensus for an autoimmune blistering disorder, which affects one or more mucous membrane and may involve the skin. The condition involves a number of different autoantigens in the basement membrane zone. It may result in severe complications from scarring, such as blindness and strictures. Diagnosis of these diseases relies on direct immunofluorescence microscopy studies and immunoserological assays. Management of affected patients is often challenging. We will here review the clinical and immunopathological features as well as the pathophysiology of this group of organ-specific autoimmune diseases. Finally, we will discuss the diagnostic approach and the principles of management in clinical practice.

Unravelling Diurnal Asymmetry of Surface Temperature in Different Climate Zones.

Understanding the evolution of Diurnal Temperature Range (DTR), which has contradicting global and regional trends, is crucial because it influences environmental and human health. Here, we analyse the regional evolution of DTR trend over different climatic zones in India using a non-stationary approach known as the Multidimensional Ensemble Empirical Mode Decomposition (MEEMD) method, to explore the generalized influence of regional climate on DTR, if any. We report a 0.36‚??¬įC increase in overall mean of DTR till 1980, however, the rate has declined since then. Further, arid deserts and warm-temperate grasslands exhibit negative DTR trends, while the west coast and sub-tropical forest in the north-east show positive trends. This transition predominantly begins with a 0.5‚??¬įC increase from the west coast and spreads with an increase of 0.25‚??¬įC per decade. These changes are more pronounced during winter and post-monsoon, especially in the arid desert and warm-temperate grasslands, the DTR decreased up to 2‚??¬įC, where the rate of increase in minimum temperature is higher than the maximum temperature. We conclude that both maximum and minimum temperature increase in response to the global climate change, however, their rates of increase are highly local and depend on the underlying climatic zone.

Direct Extraction of Excitation Energies from Ensemble Density-Functional Theory.

A very specific ensemble of ground and excited states is shown to yield an exact formula for any excitation energy as a simple correction to the energy difference between orbitals of the Kohn-Sham ground state. This alternative scheme avoids either the need to calculate many unoccupied levels as in time-dependent density functional theory (TDDFT) or the need for many self-consistent ensemble calculations. The symmetry-eigenstate Hartree-exchange (SEHX) approximation yields results comparable to standard TDDFT for atoms. With this formalism, SEHX yields approximate double excitations, which are missed by adiabatic TDDFT.

A Nose for Hydrogen Gas: Fast, Sensitive H2 Sensors Using Electrodeposited Nanomaterials.

Hydrogen gas (H2) is odorless and flammable at concentrations above 4% (v/v) in air. Sensors capable of detecting it rapidly at lower concentrations are needed to "sniff" for leaked H2 wherever it is used. Electrical H2 sensors are attractive because of their simplicity and low cost: Such sensors consist of a metal (usually palladium, Pd) resistor. Exposure to H2 causes a resistance increase, as Pd metal is converted into more resistive palladium hydride (PdHx). Sensors based upon Pd alloy films, developed in the early 1990s, were both too slow and too insensitive to meet the requirements of H2 safety sensing. In this Account, we describe the development of H2 sensors that are based upon electrodeposited nanomaterials. This story begins with the rise to prominence of nanowire-based sensors in 2001 and our demonstration that year of the first nanowire-based H2 sensor. The Pd nanowires used in these experiments were prepared by electrodepositing Pd at linear step-edge defects on a graphite electrode surface. In 2005, lithographically patterned nanowire electrodeposition (LPNE) provided the capability to pattern single Pd nanowires on dielectrics using electrodeposition. LPNE also provided control over the nanowire thickness (¬Ī1 nm) and width (¬Ī10-15%). Using single Pd nanowires, it was demonstrated in 2010 that smaller nanowires responded more rapidly to H2 exposure. Heating the nanowire using Joule self-heating (2010) also dramatically accelerated sensor response and recovery, leading to the conclusion that thermally activated H2 chemisorption and desorption of H2 were rate-limiting steps in sensor response to and recovery from H2 exposure. Platinum (Pt) nanowires, studied in 2012, showed an inverted resistance response to H2 exposure, that is, the resistance of Pt nanowires decreased instead of increased upon H2 exposure. H2 dissociatively chemisorbs at a Pt surface to form Pt-H, but in contrast to Pd, it stays on the Pt surface. Pt nanowires showed a faster response to H2 exposure than Pd nanowires operating at the same elevated temperature, but they had a surprising disadvantage: The resistance change observed for Pt nanowires was exactly the same for all H2 concentrations. Electron surface scattering was implicated in the mechanism for these sensors. Work on Pt nanowires lead in 2015 to the preparation of Pd nanowires that were electrochemically modified with thin Pt layers (Pd@Pt nanowires). Relative to Pd nanowires, Pt@Pd nanowires showed accelerated response and recovery to H2 while retaining the same high sensitivity to H2 concentration seen for sensors based upon pure Pd nanowires. A new chapter in H2 sensing (2017) involves the replacement of metal nanowires with carbon nanotube ropes decorated with electrodeposited Pd nanoparticles (NPs). Even higher sensitivity and faster sensor response and recovery are enabled by this sensor architecture. Sensor properties are strongly dependent on the size and size monodispersity of the Pd NPs, with smaller NPs yielding higher sensitivity and more rapid response/recovery. We hope the lessons learned from this science over 15 years will catalyze the development of sensors based upon electrodeposited nanomaterials for gases other than H2.

Hydrophilic, Potent, and Selective 7-Substituted 2-Aminoquinolines as Improved Human Neuronal Nitric Oxide Synthase Inhibitors.

Neuronal nitric oxide synthase (nNOS) is a target for development of antineurodegenerative agents. Most nNOS inhibitors mimic l-arginine and have poor bioavailability. 2-Aminoquinolines showed promise as bioavailable nNOS inhibitors but suffered from low human nNOS inhibition, low selectivity versus human eNOS, and significant binding to other CNS targets. We aimed to improve human nNOS potency and selectivity and reduce off-target binding by (a) truncating the original scaffold or (b) introducing a hydrophilic group to interrupt the lipophilic, promiscuous pharmacophore and promote interaction with human nNOS-specific His342. We synthesized both truncated and polar 2-aminoquinoline derivatives and assayed them against recombinant NOS enzymes. Although aniline and pyridine derivatives interact with His342, benzonitriles conferred the best rat and human nNOS inhibition. Both introduction of a hydrophobic substituent next to the cyano group and aminoquinoline methylation considerably improved isoform selectivity. Most importantly, these modifications preserved Caco-2 permeability and reduced off-target CNS binding.

Search for lepton-flavour-violating decays of the Higgs and Z bosons with the ATLAS detector.

Direct searches for lepton flavour violation in decays of the Higgs and Z bosons with the ATLAS detector at the LHC are presented. The searches are based on the data sample of proton-proton collisions collected by the ATLAS detector corresponding to an integrated luminosity of 20.3 [Formula: see text] at a centre-of-mass energy of [Formula: see text] TeV. Mutations of the recombinase-activating genes 1 and 2 (RAG1 and RAG2) in humans are associated with a broad range of phenotypes. For patients with severe clinical presentation, hematopoietic stem cell transplantation (HSCT) represents the only curative treatment; however, high rates of graft failure and incomplete immune reconstitution have been observed, especially after unconditioned haploidentical transplantation. Studies in mice have shown that Rag(-/-) natural killer (NK) cells have a mature phenotype, reduced fitness, and increased cytotoxicity. We aimed to analyze NK cell phenotype and function in patients with mutations in RAG and in non-homologous end joining (NHEJ) genes. Here, we provide evidence that NK cells from these patients have an immature phenotype, with significant expansion of CD56(bright) CD16(-/int) CD57(-) cells, yet increased degranulation and high perforin content. Correlation was observed between in vitro recombinase activity of the mutant proteins, NK cell abnormalities, and in vivo clinical phenotype. Addition of serotherapy in the conditioning regimen, with the aim of depleting the autologous NK cell compartment, may be important to facilitate engraftment and immune reconstitution in patients with RAG and NHEJ defects treated by HSCT.

Ubiquitin orchestrates proteasome dynamics between proliferation and quiescence in yeast.

Proteasomes are essential for protein degradation in proliferating cells. Little is known about proteasome functions in quiescent cells. In non-dividing yeast, a eukaryotic model of quiescence, proteasomes are depleted from the nucleus and accumulate in motile cytosolic granules termed proteasome storage granules (PSG). PSG enhance resistance to genotoxic stress and confer fitness during aging. Upon exit from quiescence PSG dissolve and proteasomes are rapidly delivered into the nucleus. To identify key players in PSG organization, we performed high-throughput imaging of green fluorescent protein (GFP)-labeled proteasomes in the yeast null mutant collection. Mutants with reduced levels of ubiquitin are impaired in PSG formation. Co-localization studies of PSG with proteins of the yeast GFP collection, mass spectrometry and direct stochastic optical reconstitution microscopy (dSTORM) of cross-linked PSG revealed that PSG are densely packed with proteasomes and contain ubiquitin but no poly-ubiquitin chains. Our results provide insight into proteasome dynamics between proliferating and quiescent yeast in response to cellular requirements for ubiquitin-dependent degradation.

Temporal Evaluation of Polybrominated Diphenyl Ether (PBDE) Serum Levels in Middle-Aged and Older California Women, 2011-2015.

In response to health concerns and widespread human exposures, two widely used commercial formulations of polybrominated diphenyl ethers (PBDEs) were banned in the United States in 2005. Initial biomonitoring data have provided early indications of reduced human exposures since these bans took effect. Our objective was to evaluate temporal trends in PBDE serum levels among a population of older California women during a four-year period, beginning approximately five years after these formulations were banned. Automated solid phase extraction and gas chromatography/high resolution mass spectrometry were used to measure PBDE levels in blood collected during 2011-2015 among 1253 women (ages 40-94) participating in the California Teachers Study. Only congeners with detection frequencies (DF) ‚Č• 75% were included in the present analysis: BDE-47 (DF = 88%); BDE-100 (DF = 78%); and BDE-153 (DF = 80%). Results from age- and race/ethnicity-adjusted linear regression analyses indicated modest, but statistically significant, average annual percent increases in the serum concentrations of all three PBDEs over the four-year study period. While not without limitations, these results, in the context of other biomonitoring data, suggest that earlier reported declines in PBDE levels may have plateaued and may now be starting to increase. Further biomonitoring to ascertain current trends and determinants of population exposures is warranted.

Time-Restricted Feeding Shifts the Skin Circadian Clock and Alters UVB-Induced DNA Damage.

The epidermis is a highly regenerative barrier protecting organisms from environmental insults, including UV radiation, the main cause of skin cancer and skin aging. Here, we show that time-restricted feeding (RF) shifts the phase and alters the amplitude of the skin circadian clock and affects the expression of approximately 10% of the skin transcriptome. Furthermore, a large number of skin-expressed genes are acutely regulated by food intake. Although the circadian clock is required for daily rhythms in DNA synthesis in epidermal progenitor cells, RF-induced shifts in clock phase do not alter the phase of DNA synthesis. However, RF alters both diurnal sensitivity to UVB-induced DNA damage and expression of the key DNA repair gene, Xpa. Together, our findings indicate regulation of skin function by time of feeding and emphasize a link between circadian rhythm, food intake, and skin health.

As the population gets older, coupled with increased awareness of good health practices and the recognition that fitness contributes to participation in activities generally appropriate for younger individuals, decisions need to be made about what are appropriate activities for the older-aged scuba (self-contained underwater breathing apparatus) diver. It is essential to appreciate the distinction between chronological and physiological age. Three factors, namely fitness, comorbidities, and mobility and strength are fundamental when making decisions about participation in activities in general as well as in scuba diving for older adults. There is almost always a time to call it quits for everything.
